Enzyme Kinetics Problem 4

Tutorial to help us answer problem 4.

 

We know from looking at the Lineweaver-Burk equation that Km/Vmaxis the slope of the line. In problem 3, we determined that the Vmax of this enzyme is 50.

Using this information, we can write,

m = Km/Vmax = Km/50 = (1/v2 - 1/v1)/(1-[s]2 - 1/[s]1.

By visually inspecting the graph we can see that points,

(1/[S]1, 1/v1) = (-0.04, 0), (1/[S]2, 1/V2) = (0, 0.02)

lie on the graph of the line. Thus, we find the slope of the line to be,

m = Km/vmax = km/500 =(0.02-0)/(0-(-0.04) = 0.5.

Solving for Km in the above equation yields,

Km = (50)(0.5) = 25 µM
